Retatrutide alleviates diabetic kidney injury via Beta-klotho-mediated ferroptosis inhibition

Retatrutide, a GIP/GLP-1/Glucagon Receptor agonist, exhibits renal protective effects in patients with diabetic kidney disease (DKD). But the exact underlying mechanism remains not fully elucidated. This study uses transcriptome sequencing and identifies 𝜷-Klotho (KLB) as the main target responsible for the role of Retatrutide in kidney protection. Retatrutide treatment decreases renal injury by inhibiting ferroptosis in patients, animal models, and HK-2 cells. Inhibition of ferroptosis by retatrutide further attenuates renal inflammation and fibrosis. This work highlights the potential of GIP/GLP-1/Glucagon Receptor agonist and KLB targeting as promising therapeutic approaches for DKD management.
